TJ proteins are necessary to maintain a strong gut barrier. When these proteins are damaged, the barrier becomes leaky. Leaky gut is associated with increased inflammation and all kinds of related health conditions.<\/p>\n\n\n\n<p>Your gut lining cells release zonulin in response to certain foods or microbes. Release of zonulin<em> reduces<\/em> <em>intestinal TJ proteins<\/em> and may cause leaky gut syndrome. It also causes inflammation and the movement of immune cells from the gut into the body. Conversely, long-term zonulin elevation may cause leaky gut. <span class=\"custom-citation\" data-id=\"ref-1\"><\/span><\/p>\n\n\n\n<p>One of the most potent triggers of zonulin is bacterial overgrowth and gut flora imbalance (dysbiosis).<span class=\"custom-citation\" data-id=\"ref-1\"><\/span><span class=\"custom-citation\" data-id=\"ref-2\"><\/span> This is one of the ways that dysbiosis contributes to leaky gut.&nbsp;<\/p>\n\n\n\n<p>Aside from dysbiosis, <a rel=\"noreferrer noopener\" href=\"https:\/\/bioptimizers.com\/blog\/the-benefits-of-a-gluten-free-diet\/\" target=\"_blank\">gluten<\/a> is the other most powerful stimulator of zonulin release.<span class=\"custom-citation\" data-id=\"ref-2\"><\/span><\/p>\n\n\n<div class=\"wp-block-image\">\n<figure class=\"aligncenter size-full\"><img loading=\"lazy\" decoding=\"async\" width=\"683\" height=\"512\" src=\"https:\/\/bioptimizers.com\/blog\/wp-content\/uploads\/2022\/08\/GettyImages-119585261-1.jpg\" alt=\"\" class=\"wp-image-150302312146\"\/><\/figure><\/div>\n\n\n<h2 class=\"wp-block-heading\">What Does High Zonulin Mean?<\/h2>\n\n\n\n<p>High zonulin levels are associated with leaky gut and therefore, many chronic inflammatory conditions.<\/p>\n\n\n\n<p>Chronic inflammation and other conditions associated with elevated zonulin levels include<span class=\"custom-citation\" data-id=\"ref-2\"><\/span>:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li><strong>Autoimmunity (multiple sclerosis, Chron\u2019s, Type 1 diabetes, etc.)<\/strong><\/li>\n\n\n\n<li>Celiac disease<\/li>\n\n\n\n<li><strong>Type 2 diabetes<\/strong><\/li>\n\n\n\n<li>Autism<\/li>\n\n\n\n<li><strong>ADHD<\/strong><\/li>\n\n\n\n<li>Depression<\/li>\n\n\n\n<li><strong>Schizophrenia<\/strong><\/li>\n\n\n\n<li>Irritable bowel syndrome (IBS)<\/li>\n<\/ul>\n\n\n\n<p>Testing for zonulin however, is not perfect and does not always correlate with the presence of leaky gut or disease.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Problems With Zonulin Testing&nbsp;<\/h2>\n\n\n\n<p>Zonulin was first discovered as a gut response to the bacteria <em>Vibrio cholera<\/em>. A zonulin test called enzyme-linked immunosorbent assay (ELISA) was developed. ELISA testing is a method where a tray is used that has molecules in it that recognizes and sticks to the substance being tested.<\/p>\n\n\n\n<p>The lab sample is placed in the tray. A machine then calculates how much of the substance, in this case zonulin, is present based on how much binds to the ELISA molecule. The trouble with zonulin ELISA testing is that the original \u201csticky\u201d molecule developed doesn\u2019t effectively measure zonulin.&nbsp;<\/p>\n\n\n\n<p>In short, zonulin tests currently available don\u2019t appear to actually measure zonulin. They measure something related to zonulin, like a precursor or another related molecule.<span class=\"custom-citation\" data-id=\"ref-3\"><\/span> Scientists are still trying to figure that part out.<\/p>\n\n\n\n<p>Testing for zonulin may be helpful as part of a broader plan to examine gut health, but it is not always enough to diagnose leaky gut on its own. The most likely contributors to elevated zonulin are gut flora imbalance and food sensitivities.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Balance Dysbiosis<\/h3>\n\n\n\n<p><a href=\"https:\/\/bioptimizers.com\/blog\/reset-your-gut-in-3-simple-steps\/\" target=\"_blank\" rel=\"noreferrer noopener\">Dysbiosis<\/a> triggers inflammation and zonulin release from gut cells.
